Competitive enzymatic reaction to control allele-specific extensions
Here, we present a novel method for SNP genotyping based on protease-mediated allele-specific primer extension (PrASE), where the two allele-specific extension primers only differ in their 3'-positions. As reported previously [Ahmadian,A., Gharizadeh,B., O'Meara,D., Odeberg,J. and Lundeberg,J. (2001), Nucleic Acids Res., 29, e121], the kinetics of perfectly matched primer extension is faster th...
متن کاملHaptoglobin genotyping by allele-specific polymerase chain reaction amplification.
We performed haptoglobin (Hp) genotyping by polymerase chain reaction using allele-specific primer-pairs. The major six genotypes of Hp were identified using this method. Among Japanese individuals living in Ehime and Okayama Prefectures, the allele frequencies were estimated to be Hp2 = 0.723 and Hp1s = 0.277. Genotyping of Hp was possible with 0.3 ng of DNA and with 0.125 microliter of blood....
متن کاملallele-specific polymerase chain reaction for detection of main gyra allelic variants in helicobacter pylori strains
background rapid detection of resistant strains of helicobacter pylori in human clinical samples is of major importance in clinical settings. inability of conventional clinical laboratory techniques in detection of these strains usually leads to failure of prescribed therapeutic regimens. objectives the aim of this study was designing a simple and rapid allele-specific pcr (as-pcr)-based method...
متن کاملCellular Automata Approaches to Enzymatic Reaction Networks
Cellular automata simulations for enzymatic reaction networks differ from other models for reaction-diffusion systems, since enzymes and metabolites have very different properties. This paper presents a model where each lattice site can can contain at most one enzyme molecule, but many metabolite molecules. The rules are constructed to conform to the Michaelis-Menten kinetics by modeling the un...
متن کاملPolymorphism of rs2076530 allele in BTNL2 gene in patients with skin sarcoidosis compared to skin sarcoidal reaction and normal skin: A case-control study
Background: In 10-30% of the cases with sarcoidosis, skin lesions appear solely without any systemic signs or symptoms. BTNL2 gene, which is a member of the immunoglobulin gene super family and is associated with CD86 and CD80 co-stimulatory receptors, is identified to play an important role in the establishment of sarcoidosis. We aimed to evaluate the role of this gene in patients with skin sa...
